The standard says "customarily and regularly" which is defined as "greater than occasional but less than constant; it includes work normally done every workweek, but does not include isolated or one-time tasks." You are correct, overtime is owed for hours worked in one week over 40. Keep in mind that they are free to set the day the work week begins according to their needs; it need not be on the weekend.
